Redox-active molecules can diffuse inside cells and across cell membrane, and are increasingly recognized for their important functions in cell signaling. We have recently utilized genetically encoded unnatural amino acids and fluorescent proteins to develop encodable fluorescent probes for redox-active molecules. We have derived highly selective probes and introduced them into mammalian cells to monitor redox dynamics. We are also integrating these fluorescent probes with other bioanalytical methods to study mitochondrial oxidative/nitrosative stress under various pathophysiological conditions.
